1.凍結切片のための組織調製

</ol>2.凍結切片
<ol><li>切片の準備ができたら、組織の粉砕を防ぐために、切片化の前に少なくとも1〜2時間クライオスタットでサンプルを順応させます。</li>
	<li>クライオスタットを使用して、組織を切片(20〜50 μm)に切断し、1x PBS溶液で満たされた6または12ウェルインサート( 材料の表を参照)に収集します。 注:セクションの厚さ、収集する組織の量、および使用するウェルインサートの数に応じて、各ウェルには、ウェル用に約10〜40スライスにまたがる可変数のセクションが含まれます。たとえば、脳全体を40μmで切片化する場合、12ウェルインサートを使用して各ウェルに約18〜24の切片が収集されます。また、20 μmの切片は取り扱いがやや難しい場合があるため、バルク染色には40 μmをお勧めします(「説明」を参照)。</li>
</ol>3. セクションの保存
<ol><li>採取したら、作りたての1x PBSで切片を5分間洗浄します。3回繰り返します。</li>
	<li>切片を1〜1.5 mLの保存溶液で満たされた2 mLの微量遠心チューブに移します(250 mLの場合、70 gのスクロース、75 mLのエチレングリコールを混合し、0.1 Mリン酸バッファーで容量にします)。</li>
	<li>染色の準備ができるまで-80°Cで保存してください。</li>
</ol>4.染色I日目
<ol><li>冷凍庫からサンプルを取り出し、室温(RT)で10〜20分間平衡化します。</li>
	<li>セクションを6ウェルプレートのウェルインサートに注ぎ、保存液をセクションから分離します。</li>
	<li>ウェルインサートを、約6 mLの1x TBSを含む別のウェルに移動します。 RTで低速で低速を使用して、オービタルシェーカーで1x TBSでそれぞれ5分間3回洗浄します。</li>
	<li>切片を洗浄している間に、0.3%Triton X-100を含む1x TBSと3%正常血清(例:.、正常ウマ血清)からなるブロッキング透過液7 mL(サンプルあたり)を調製します。低速を使用して、オービタルシェーカーのRTで30分間セクションをブロックします。 注:血清によるブロッキングは、組織または非特異的Fc受容体への抗体の非特異的結合を防ぎます–二次抗体の種と一致する血清が推奨されますが、利用できない場合は、一次抗体宿主動物とは異なる種からの任意の正常血清を使用できます。界面活性剤Triton X-100は、組織を透過させることにより、抗体の浸透を改善します。</li>
	<li>選択した一次抗体(適切に希釈)からなる一次抗体溶液のサンプルあたり1 mLを、0.3%Triton X-100および1%正常血清を含む1x TBSで調製します(ステップ4.4注を参照)。ウェルインサートから一次抗体溶液を含む2 mLマイクロ遠心チューブに切片を移し、目的の抗原に結合させます。 注:複数の一次抗体を使用できます(異なる宿主種で生成)。</li>
	<li>切片付きの2 mLマイクロ遠心チューブを低速(速度7 rpmなど)を使用して回転ミキサーに置き、4°Cで12〜16時間一晩インキュベートします。</li>
</ol>5.染色2日目
<ol><li>翌日、切片を6ウェルプレートのウェルインサートに注ぎ、一次抗体溶液から切片を分離します。 注:抗体溶液は収集して再利用できます。微生物の増殖を抑制するために0.02%(w / v)アジ化ナトリウムを追加します。</li>
	<li>RTで1x TBSでセクションを3回洗浄します(最初の2回の洗浄で30秒、最後の洗浄で10分)。</li>
	<li>0.3%Triton X-100および1%正常血清(光からの遮蔽液)を含む1x TBS中の適切な二次抗体(それに応じて希釈)からなる二次抗体溶液をサンプルあたり1 mL調製します。 注:結合二次抗体による間接標識は、シグナルを増幅し、タンパク質標的の比色または蛍光可視化を可能にします。</li>
	<li>二次抗体溶液を入れた2 mLの微量遠心チューブに切片を移します。低速(光からの遮蔽溶液)を使用して、オービタルシェーカーのRTで2時間インキュベートします。</li>
	<li>切片を6ウェルプレートのウェルインサートに注ぎ、二次抗体溶液から切片を分離します。</li>

</ol>6. 取り付け
<ol><li>1x TBSで4分の3を満たしたガラス製の長方形の組織学的チャンバーに切片を注ぎます。</li>

</ol>7.カバースリップ

免疫組織化学(IHC)は、組織切片内のタンパク質発現と局在を特定する上で重要になっている汎用性の高い技術です。このアッセイは、正常な機能の段階から病状までの組織の特徴をさらに理解するために、科学界全体で使用されています。IHCは、がんなどの疾患の臨床診断から前臨床研究における最初の発見まで、さまざまな分野で採用されています10,36。
IHCを実行するために最も一般的に使用される技術は、切片がスライスされた後すぐにスライドに接着されるスライドマウント法です。この技術を使用するいくつかの利点は、研究者がタンパク質共局在研究に必要な非常に薄い切片を処理でき、スライドごとに切片を染色するための溶液をほとんど使用しないことです。抗体はしばしば高価です。したがって、このアプローチは、処理するセクションが少ない場合に経済的なオプションになる可能性があります。これは、組織の取り扱いが最小限であり、組織の構造的完全性が保護されるため、新鮮凍結標本を使用する研究者にも最適な方法です。スライドマウントアプローチを使用することは、臨床病理学の場合のように、少数の切片のみを採取してすぐに染色する場合にも適切です。一方、染色中に組織の露出側のみにアクセスし、抗体の浸透が不十分で効果的な洗浄のために切片の厚さが制限されるなどの欠点があります。別の欠点は、組織が切片化されてスライドに集められると、IHCは通常、未処理のスライドの保管が多くの冷凍庫スペースを占めるため、かなり迅速に完了する必要があることです。さらに、より大きな実験(例えば、染色されるべき複数の代表的なレベルを有する複数の脳領域)を処理する場合、このアプローチは実際にはより多くの試薬を使用する可能性があり、かなり時間がかかり、実験ごとに処理されるスライドの数を制限することができることが多い。
スライドマウント法に関連するいくつかの制限は、より厚い切片を扱うときにますます普及している代替手段となっているフリーフローティング染色技術によって克服できます。この方法は新しいアプローチではありませんが、私たちの経験では、特に組織サンプルを大量に染色する場合、信頼性が高く、再現性があり、柔軟なアプローチであるため、より大規模な研究を効率的に処理できます。研究者は、この方法を使用して、複数の大規模なIHC実験を同時に効果的に実行することもできます。さらに、サンプルは懸濁液で染色されるため、溶液はあらゆる角度から切片に浸透する可能性があり、特に厚い切片にとって重要であり、多くの場合、より高品質の染色につながります(図2、図3、 および 図5)。フリーフローティング切片は、厚さ37で20〜50μmの範囲でスライスでき、より厚い切片は、研究者がさまざまな視野で構造や細胞を見るのに役立ちます。たとえば、脳組織では、より厚い切片により、研究者はサンプル全体の樹状突起と軸索の構造を見ることができます。より薄いスライス(20μm)を収集する機能により、アプリケーションのスペクトルがさらに広がります。ただし、薄いスライスは取り扱いが難しく、損傷を最小限に抑えるためにより多くの時間と労力が必要になることが多いため、バルク染色のために切片を40μmより薄くしないでください。フリーフローティング法の主な利点の1つは、研究者が脳全体(または他の組織)をすばやく切片化し、すべての切片を小さなチューブに集め、各アリコートがすべての異なる脳領域の代表的なスライスを持つため、研究者が脳全体を素早く染色できることです。スライスを含むチューブは、-80°Cの凍結保護剤に数年間保存することができ33、アリコートは冷凍庫のスペースをあまり占有しないため、研究者は効果的に「組織ライブラリ」を生成できます。この方法はまた、スライド、カバーガラス、特に再利用のために簡単に回収して保存できる抗体や、ユーザーが選択する限り切片を保存および保存できるため、貴重な動物組織などの無駄な材料の量を削減します。
ここで紹介するフリーフローティングアプローチとプロトコルは、研究者にプロトコルを簡単に変更したり、リソースを再利用したりするオプションも提供します。例えば、採取した切片は、発色性IHC、ヘマトキシリンおよびエオシン(H&E)、クレジルバイオレット(図4)、RNAscope38などの単純なプロトコル修飾による免疫蛍光に加えて、多くの異なる組織化学的染色に使用できます。発色IHCは、可溶性基質が二次抗体に結合した酵素によって不溶性発色産物に変換された場合の抗原発現の可視化を可能にします。最も一般的に使用される2つの酵素は、3,3'ジアミノベンジジン(DAB)を暗褐色の最終生成物に変換する西洋ワサビペルオキシダーゼ(HRP)と、3-アミノ-9-エチルカルバゾール(AEC)基質を赤色生成物に変換するアルカリホスファターゼ(AP)です39。私たちは日常的にクレシルバイオレット染色を行い、脳の肉眼組織と形態を調べるために浮遊切片を使用しています40。また、このプロトコルを脳、肝臓、心臓、腎臓、脾臓など、さまざまな組織に適用することも成功しています(図5)。他の研究者も、肝臓、腎臓、卵巣などの末梢組織にこの手法を使用することに成功しています22,23,24。
フリーフローティング技術を使用する場合の主な懸念は、プロトコル全体を通して、サンプルが均一に洗浄、ブロック、および染色されるように、ほぼすべてのステップでシェーカーとローテーター上にあるため、組織切片、特に脳スライスの構造的損傷の可能性です。時折、特定の脳領域は、特に小脳レベルで剥離することがあります。ただし、取り付けプロセス中に脳アトラスと宝石商のランプなどの拡大の形式を使用すると、セクションをつなぎ合わせるのに役立ちます。この課題は通常、サンプルを穏やかに取り扱い、回転機械を正しい低い設定に保つことで防ぐことができます。
結論として、さまざまな組織のタンパク質発現と局在、および組織構造を視覚化するために定期的に使用する、不可欠で信頼性が高く、柔軟で効率的なモダリティであることが証明されている、確立されたフリーフローティングIHC技術を紹介します。ここに記載されているプロトコルは、個々の研究ニーズに合わせて簡単に変更できるため、科学界にとって価値があります。

免疫染色は、130年前の1890年にフォンベーリング1によって血清抗体が発見されたことから始まった人気のある研究慣行です。20世紀初頭、反応を定量および視覚化する方法として色素が抗原に結合し、後に抗体に付着し1、1941年にアルバートクーンズは最初の蛍光抗体標識を開発し、光学顕微鏡法に革命をもたらした発見2,3。「免疫染色」という用語は、ウェスタンブロット、フローサイトメトリー、ELISA、免疫細胞化学、および免疫組織化学を含む、この原理を用いて開発された多くの技術を包含する3,4。ウェスタンブロットは、組織または細胞抽出物から特定のタンパク質の存在を検出します5.タンパク質は、ゲル電気泳動を使用してサイズ別に分離され、メンブレンに転写され、抗体を使用してプローブされます。この手法は、タンパク質の存在とタンパク質が存在する量を示します。しかしながら、それは細胞または組織内のタンパク質の局在に関する情報を明らかにしない。別の方法である免疫細胞化学(ICC)は、細胞内のタンパク質、典型的にはインビトロで培養された細胞を標識する。ICCは、細胞コンパートメント内のタンパク質発現と局在の両方を示します6。特定のタンパク質を組織レベルで検出および視覚化するために、免疫組織化学(IHC)が利用されます。
IHCは、免疫系の化学的性質を利用して、組織内の特定の抗原を標的とするために研究者が使用する方法です7,8。酵素または蛍光色素のいずれかに結合した特異的な一次抗体および二次抗体を生成することにより、目的の抗原を標識し、ほとんどの組織で明らかにすることができます(Mepham and Brittenでレビューされています)9。「免疫組織化学」という用語自体は、目的の抗原を明らかにするために使用される標識方法を指定しません。したがって、この用語は、標識方法を明確に描写するために検出技術と組み合わせることがよくあります:二次抗体がペルオキシダーゼなどの酵素に結合していることを示す発色免疫組織化学(CIH)。または蛍光IHCは、二次抗体がフルオレセインイソチオシアネート(FITC)またはテトラメチルローダミン(TRITC)などのフルオロフォアに結合している場合を示します。IHCの選択性により、臨床医や研究者は、さまざまな健康状態や疾患状態にわたる組織全体のタンパク質の発現と分布を視覚化できます10。臨床分野では、IHCは一般的に癌の診断やさまざまな種類の癌の違いを判断するために使用されます。IHCは、B型肝炎やC11型肝炎など、体内のさまざまな種類の微生物感染を確認するためにも使用されています。生物医学研究では、IHCは組織内のタンパク質発現をマッピングするためによく使用され、病状に見られる異常なタンパク質を特定するのに重要です。例えば、神経変性は、アルツハイマー病におけるΑβプラークや神経原線維変化などの異常なタンパク質の脳内の蓄積を伴うことが多い。動物モデルは、これらの病理学的状態を模倣するために開発されることが多く、IHCは、研究者が目的のタンパク質を見つけて定量するために使用する1つの方法です10、12、13。次に、これらの病気の原因とそれらに伴って発生する合併症についてもっと学ぶことができます。
IHCの実行には多くのステップがあります。まず、目的の組織を回収し、染色の準備をします。おそらくほとんどの研究者は固定組織サンプルを準備し、循環器系を介した固定液の灌流は形態を保存するため最適です14,15。組織サンプルの事後固定も使用され得るが、理想的よりも低い結果をもたらす可能性がある16。ホルムアルデヒドなどの架橋固定剤は、組織17内のタンパク質間に化学結合を作り出すことによって作用する。次に、固定された組織はミクロトームを使用して非常に薄い層または切片にスライスされ、多くの研究者はクライオスタットを使用して凍結切片を収集することを好みます。そこから組織を採取し、顕微鏡スライドに直接取り付けるか(スライドマウント法)、リン酸緩衝生理食塩水(PBS)などの溶液に懸濁します(フリーフローティング法)18。使用される収集方法は、研究者のニーズに基づいて事前に決定されており、これら2つの方法のそれぞれが独自の長所と短所を示しています。
スライドマウント法は群を抜いて最も一般的に使用されており、非常に薄い切片(10〜14μm)を調製できることが重要な利点であり、これは例えばタンパク質間相互作用を調べるために重要です。標本の取り扱いも最小限であり、これは組織19の構造的完全性に対する潜在的な損傷を減少させる。研究者は、固定組織と比較して非常にデリケートであり、サンプルの解凍を防ぐために多くの注意を払う必要がある新鮮な凍結組織(ドライアイス、イソペンタンなどを使用してすぐに凍結される組織)でこの技術を使用することがよくあります。スライドマウント切片を使用するもう一つの利点は、染色のための大量の溶液が通常必要とされないことです4。したがって、研究者は少量の高価な抗体または他の化学物質を使用して染色を完成させることができます。さらに、複数の異なる実験グループのセクションを同じスライドにマウントすることが可能であり、特に画像取得時に有利です。一方、スライドマウント切片を使用することにはいくつかの欠点があり、最も顕著なのは、組織切片がスライドに接着されているため、抗体の浸透が切片の片側に制限され、切片の厚さと組織の3D表現が制限されることです。また、洗浄中に、組織の端とセクション全体がスライドから剥がれ、実験全体が役に立たなくなることもあります。さらに、IHCは通常、抗原エピトープ20,21の分解を回避するためにスライドマウントアプローチを使用する場合、比較的迅速に実行する必要があり、未処理のスライドは通常-20または-80°Cで保存され、多くの場合、カバースリップして水平またはスライドボックスに保存され、比較的大きな保存フットプリントになります。最後に、研究者が多数の組織切片を処理するために多数のスライドを処理する必要がある場合、スライドマウント技術は時間がかかる可能性もあります。

Techniques for Processing Eyes Implanted With a Retinal Prosthesis for Localized Histopathological Analysis

With the recent development of retinal prostheses, it is important to develop reliable techniques for assessing the safety of these devices in preclinical studies. However, the standard fixation, preparation, and automated histology procedures are not ideal. Here we describe new procedures for evaluating the health of the retina directly adjacent to an implant. Retinal prostheses feature electrode arrays in contact with eye tissue. Previous methods have not been able to spatially localize the ocular tissue adjacent to individual electrodes within the array. In addition, standard histological processing often results in gross artifactual detachment of the retinal layers when assessing implanted eyes. Consequently, it has been difficult to assess localized damage, if present, caused by implantation and stimulation of an implanted electrode array. Therefore, we developed a method for identifying and localizing the ocular tissue adjacent to implanted electrodes using a (color-coded) dye marking scheme, and we modified an eye fixation technique to minimize artifactual retinal detachment. This method also rendered the sclera translucent, enabling localization of individual electrodes and specific parts of an implant. Finally, we used a matched control to increase the power of the histopathological assessments. In summary, this method enables reliable and efficient discrimination and assessment of the retinal cytoarchitecture in an implanted eye.

Primer for Immunohistochemistry on Cryosectioned Rat Brain Tissue: Example Staining for Microglia and Neurons

Immunohistochemistry is a widely used technique for detecting the presence, location, and relative abundance of antigens in situ. This introductory level protocol describes the reagents, equipment, and techniques required to complete immunohistochemical staining of rodent brain tissue, using markers for microglia and neuronal elements as an example. Specifically, this paper is a step-by-step protocol for fluorescent visualization of microglia and neurons via immunohistochemistry for Iba1 and Pan-neuronal, respectively. Fluorescence double-labeling is particularly useful for the localization of multiple proteins within the same sample, providing the opportunity to accurately observe interactions between cell types, receptors, ligands, and/or the extracellular matrix in relation to one another as well as protein co-localization within a single cell. Unlike other visualization techniques, fluorescence immunohistochemistry staining intensity may decrease in the weeks to months following staining, unless appropriate precautions are taken. Despite this limitation, in many applications fluorescence double-labeling is preferred over alternatives such as 3,3&#39;-diaminobenzidine tetrahydrochloride (DAB) or alkaline phosphatase (AP), as fluorescence is more time efficient and allows for more precise differentiation between two or more markers. The discussion includes troubleshooting tips and advice to promote success.

Highly-Multiplexed Tissue Imaging with Raman Dyes

Visualizing a vast scope of specific biomarkers in tissues plays a vital role in exploring the intricate organizations of complex biological systems. Hence, highly multiplexed imaging technologies have been increasingly appreciated. Here, we describe an emerging platform of highly-multiplexed vibrational imaging of specific proteins with comparable sensitivity to standard immunofluorescence via electronic pre-resonance stimulated Raman scattering (epr-SRS) imaging of rainbow-like Raman dyes. This method circumvents the limit of spectrally-resolvable channels in conventional immunofluorescence and provides a one-shot optical approach to interrogate multiple markers in tissues with subcellular resolution. It is generally compatible with standard tissue preparations, including paraformaldehyde-fixed tissues, frozen tissues, and formalin-fixed paraffin-embedded (FFPE) human tissues. We envisage this platform will provide a more comprehensive picture of protein interactions of biological specimens, particularly for thick intact tissues. This protocol provides the workflow from antibody preparation to tissue sample staining, to SRS microscope assembly, to epr-SRS tissue imaging.

A Rapid Method for Multispectral Fluorescence Imaging of Frozen Tissue Sections

Multispectral fluorescence imaging on formalin-fixed paraffin-embedded (FFPE) tissues enables the detection of multiple markers in a single tissue sample that can provide information about antigen coexpression and spatial distribution of the markers. However, a lack of suitable antibodies for formalin-fixed tissues may restrict the nature of markers that can be detected. In addition, the staining method is time-consuming. Here we describe a rapid method to perform multispectral fluorescence imaging on frozen tissues. The method includes the fluorophore combinations used, detailed steps for the staining of mouse and human frozen tissues, and the scanning, acquisition, and analysis procedures. For staining analysis, a commercially available semiautomated multispectral fluorescence imaging system is used. Through this method, up to six different markers were stained and detected in a single frozen tissue section. The machine learning analysis software can phenotype cells that can be used for quantitative analysis. The method described here for frozen tissues is useful for the detection of markers that cannot be detected in FFPE tissues or for which antibodies are not available for FFPE tissues.

Short-Term Free-Floating Slice Cultures from the Adult Human Brain

Organotypic, or slice cultures, have been widely employed to model aspects of the central nervous system functioning in vitro. Despite the potential of slice cultures in neuroscience, studies using adult nervous tissue to prepare such cultures are still scarce, particularly those from human subjects. The use of adult human tissue to prepare slice cultures is particularly attractive to enhance the understanding of human neuropathologies, as they hold unique properties typical of the mature human brain lacking in slices produced from rodent (usually neonatal) nervous tissue. This protocol describes how to use brain tissue collected from living human donors submitted to resective brain surgery to prepare short-term, free-floating slice cultures. Procedures to maintain and perform biochemical and cell biology assays using these cultures are also presented. Representative results demonstrate that the typical human cortical lamination is preserved in slices after 4 days in vitro (DIV4), with expected presence of the main neural cell types. Moreover, slices at DIV4 undergo robust cell death when challenged with a toxic stimulus (H2O2), indicating the potential of this model to serve as a platform in cell death assays. This method, a simpler and cost-effective alternative to the widely used protocol using membrane inserts, is mainly recommended for running short-term assays aimed to unravel mechanisms of neurodegeneration behind age-associated brain diseases. Finally, although the protocol is devoted to using cortical tissue collected from patients submitted to surgical treatment of pharmacoresistant temporal lobe epilepsy, it is argued that tissue collected from other brain regions/conditions should also be considered as sources to produce similar free-floating slice cultures.

A protocol to prepare free-floating slice cultures from adult human brain is presented. The protocol is a variation of the widely used slice culture method using membrane inserts. It is simple, cost-effective, and recommended for running short-term assays aimed to unravel mechanisms of neurodegeneration behind age-associated brain diseases.

Free-floating Immunostaining of Mouse Brains

Immunohistochemical staining of mouse brains is a routine technique commonly used in neuroscience to investigate central mechanisms underlying the regulation of energy metabolism and other neurobiological processes. However, the quality, reliability, and reproducibility of brain histology results may vary among laboratories. For each staining experiment, it is necessary to optimize the key procedures based on differences in species, tissues, targeted proteins, and the working conditions of the reagents. This paper demonstrates a reliable workflow in detail, including intra-aortic perfusion, brain sectioning, free-floating immunostaining, tissue mounting, and imaging, which can be followed easily by researchers in this field.
Also discussed are how to modify these procedures to satisfy the individual needs of researchers. To illustrate the reliability and efficiency of this protocol, perineuronal nets were stained with biotin-labeled Wisteria florbunda agglutinin (WFA) and arginine vasopressin (AVP) with an anti-AVP antibody in the mouse brain. Finally, the critical details for the entire procedure have been addressed, and the advantages of this protocol compared to those of other protocols. Taken together, this paper presents an optimized protocol for free-floating immunostaining of mouse brain tissue. Following this protocol makes this process easier for both junior and senior scientists to improve the quality, reliability, and reproducibility of immunostaining studies.

Staining and High-Resolution Imaging of Three-Dimensional Organoid and Spheroid Models

In vitro three-dimensional (3D) cell culture models, such as organoids and spheroids, are valuable tools for many applications including development and disease modeling, drug discovery, and regenerative medicine. To fully exploit these models, it is crucial to study them at cellular and subcellular levels. However, characterizing such in vitro 3D cell culture models can be technically challenging and requires specific expertise to perform effective analyses. Here, this paper provides detailed, robust, and complementary protocols to perform staining and subcellular resolution imaging of fixed in vitro 3D cell culture models ranging from 100 &#181;m to several millimeters. These protocols are applicable to a wide variety of organoids and spheroids that differ in their cell-of-origin, morphology, and culture conditions. From 3D structure harvesting to image analysis, these protocols can be completed within 4-5 days. Briefly, 3D structures are collected, fixed, and can then be processed either through paraffin-embedding and histological/immunohistochemical staining, or directly immunolabeled and prepared for optical clearing and 3D reconstruction (200 &#181;m depth) by confocal microscopy.

Here, we provide detailed, robust, and complementary protocols to perform staining and subcellular resolution imaging of fixed three-dimensional cell culture models ranging from 100 &#181;m to several millimeters, thus enabling the visualization of their morphology, cell-type composition, and interactions.

Histological Staining of Neural Tissue

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections. In the study of neuroanatomy, histological techniques are frequently applied to visualize and study nervous system tissue. This video focuses on histological staining techniques for neural tissue. An overview of common brain stains is provided, including those that specifically mark neuronal cell bodies, like Nissl stains, and those that selectively highlight myelinated axons, like the Luxol Fast blue stain. Immunohistological techniques, which take advantage of the specific interaction between antibodies and unique cellular proteins, are also discussed. Next, the preparation of brain samples for staining is described, including the basic steps for fixation, embedding, sectioning, and rehydration of the tissue. The presentation also provides a step-by-step procedure for immunohistological staining followed by a Nissl stain, in addition to practical applications of these techniques.

Histological Sample Preparation for Light Microscopy

Histology is the study of cells and tissues, which is typically aided by the use of a light microscope. The preparation of histological samples can vary greatly based on the inherent properties of the samples such as size and hardness as well as expected post-processing which includes planned staining techniques or other down-stream applications. As described in this video, specimen preparation typically begins with a fixation procedure to prevent degradation of the sample by naturally occurring enzymes that are released by the cells upon death. Once fixed, samples are placed into an embedding medium that is able to sufficiently support the sample. Most commonly this is paraffin wax, but other materials such as a glycerin based freezing medium and agars are also used to surround the sample during sectioning. Sectioning then takes place on a microtome or other cutting device that allows the user to shave the sample into thin slices ranging from a few microns to a few millimeters in thickness. Once cut, sections are mounted on a glass slide and stained to bring out specific features of the sample before being imaged on a microscope.
